Khumalo said Foreign Minister Nkosazana Dlamini-Zuma is accompanying Mbeki to the meeting.
Several senior government officials form part of the South African delegation.
Nigerian President Olusegun Obasanjo and Abdoulaye Wade, the President of Senegal, would attend the meeting, the Foreign Affairs Department said earlier.
Mbeki is expected to address the Ticad III gathering this morning, and his speech would focus on how to advance the New Partnership for Africa's Development (Nepad) through Ticad.
"The president is also going to have bilateral discussions with the Prime Minister of Japan, Koizumi. They are going to have the bilateral discussions on the margins of the Ticad," Khumalo said.
He said Mbeki would meet top officials from Japanese vehicle manufacturer Mitsubishi before his return to South Africa tomorrow.
Mitsubishi is the 10th largest company in the world, and trade between South Africa and Japan amounted to R44-billion in 2000, Khumalo said.
